化驗程序

細胞遷移實驗。使用Oris的細胞遷移實驗。™細胞遷移檢測試劑盒（Platypus Technologies）與Kui Lin的實驗室（Genentech）合作，使用PC-3細胞模型系統(tǒng)進行細胞遷移檢測。簡而言之，將細胞以15,000個細胞/孔的密度接種，形成單層。從每個孔中移除細胞接種塞，用PBS沖洗一次以去除任何未粘附的細胞。加入培養(yǎng)基和/或藥物，孵育24小時。加入在PBS中制備的a鈣蛋白aμ熒光素(最終濃度0.5μμ)標記細胞，在ImageXpress Velos系統(tǒng)上進行熒光成像。

細胞遷移的熒光成像用于此演示的ImageXpress Velos系統(tǒng)-DL激光掃描平臺配備了20mW488nm激光器。488nm的激光器用于使用通道1(Ch1)510-540nm帶通濾光片檢測鈣黃綠素標記的細胞。圖像采集在5x5微米采樣和整個96孔板進行。
