3.5 細胞的接種和培養(yǎng) 參照文獻進行。
3.6 精原細胞的純化及其純度評估：分離細胞接種后，定時觀察貼壁情況。當見到混雜其中的睪丸體細胞開始貼壁時，小心傾斜培養(yǎng)皿，將尚未貼壁的精原細胞連同培養(yǎng)液一起吸出，另行接種培養(yǎng)。待細胞貼壁后(大約培養(yǎng)12～24h)，棄去舊液，用PBS洗1～2次，加入新鮮培養(yǎng)液。任選3個視野，用計數(shù)器計每個視野中的細胞總數(shù)和精原細胞數(shù)(圓形的精原細胞附著在多突起的支持細胞上，呈鏈狀、葡萄串狀或以散在方式存在)，然后分別計算均值±標準差(0.gif (881 bytes)±s)。
4.電鏡樣品的制備
將新分離后位于27%～35% Percoll梯度間的細胞離心成團，棄去培養(yǎng)液，加入2ml 2.5%戊二醛，4℃下預固定24h；PBS清洗后轉(zhuǎn)至1%的鋨酸中后固定2h，逐級丙酮脫水，Epon812包埋，超薄切片機切片，JEM-1 200EX Ⅱ電鏡觀察(分布在19%～27%及35%～43%Percoll梯度間的細胞太少，沒有制成電鏡樣品)。
結(jié)果
1.生精上皮的消化
4次消化所獲懸液量均為1.5ml。如表3所示，平均每10個睪丸可獲得4.219×106個細胞；其中活細胞、死細胞、細胞團平均所占百分比依次為90.08%、9.92%和8.91%；平均每個睪丸可獲得4.136×105個細胞。
2.生精上皮細胞在Percoll梯度中的分布

1. Digestion of germinal epithelium
