描述：
pGEM® -T Easy 載體系統(tǒng)是克隆PCR 產(chǎn)物的方便系統(tǒng)。它除了具有pGEM® -T 載體系統(tǒng)(Cat.# A3600,A3610) 所有的優(yōu)勢外，在插入位點兩側(cè)分別有EcoRI 和NotI 酶切位點，便于通過選擇單酶切釋放插入的DNA。pGEM® -T Easy載體系統(tǒng)II 除了pGEM® -T Easy載體系統(tǒng)I 所有成分外，還包括JM109 感受態(tài)細(xì)胞。
特點:
靈活：pGEM®-T Easy 載體多克隆位點兩側(cè)分別有BstZI, EcoRI和NotI 酶切位點，便于通過選擇單酶切釋放插入的DNA。
快速連接：使用2X 快速連接緩沖液(2X Rapid Ligation Buffer)可以使連接反應(yīng)在室溫下1 小時內(nèi)完成。
藍(lán)/ 白篩選：載體包含有T7 和SP6 RNA 聚合酶啟動子，分別位于β - 半乳糖苷酶的α - 肽編碼區(qū)內(nèi)多克隆位點的兩側(cè)。α - 肽的插入失活允許在指示培養(yǎng)基上用顏色直接篩選重組克隆。

pGEM®-T和pGEM®-T Easy 載體系統(tǒng)可用于PCR產(chǎn)物的克隆。這兩種載體是通過 EcoRⅤ酶切pGEM®-5Zf(+)和pGEM® -T Easy載體，并在3’末端加入胸腺嘧啶構(gòu)建的。插入位點3’-T突出端可提高PCR產(chǎn)物的連接效率，因為3’-T突出端可以防止載體的自身環(huán)化，并且為熱穩(wěn)定性聚合酶產(chǎn)生PCR產(chǎn)物提供匹配堿基。這些聚合酶常常以不依賴模板方式在擴增產(chǎn)物的3’端加上一個脫氧腺苷酸混合物。 pGEM®-T和pGEM®-T Easy高拷貝數(shù)載體包含有T7和SP6 RNA聚合酶啟動子，其側(cè)翼和多克隆位點區(qū)相接，多克隆位點區(qū)位于β半乳糖苷酶的α肽編碼區(qū)內(nèi)。α肽插入失活允許在指示培養(yǎng)基用顏色直接篩選重組克隆。另外這兩個載體的多克隆位點區(qū)的限制性酶切位點適用于采用Promega公司Erase-a-Base®系統(tǒng)（產(chǎn)品目錄號#E5750）產(chǎn)生一系列巢式缺失體。

The pGEM®-T and pGEM®-T Easy Vector Systems are convenient systems for the cloning of PCR products. The vectors are prepared by cutting the pGEM®-5Zf(+) and pGEM®-T Easy Vectors, respectively, with EcoR V and adding a 3´ terminal thymidine to both ends. These single 3´-T overhangs at the insertion site greatly improve the efficiency of ligation of a PCR product into the plasmids by preventing recircularization of the vector and providing a compatible overhang for PCR products generated by certain thermostable polymerases. These polymerases often add a single deoxyadenosine, in a template-independent fashion, to the 3´-ends of the amplified fragments.
The high copy number pGEM®-T and pGEM®-T Easy Vectors contain T7 and SP6 RNA polymerase promoters flanking a multiple cloning region within the alpha-peptide coding region of the enzyme beta-galactosidase. Insertional inactivation of the alpha-peptide allows recombinant clones to be directly identified by blue/white screening on indicator plates.
The pGEM®-T and pGEM®-T Easy Vector Systems include a 2X Rapid Ligation Buffer for ligation of PCR products. Reactions using this buffer may be incubated for 1 hour at room temperature. The incubation period may be extended to increase the number of colonies after transformation.
注意事項：
1． 在使用pGEM®-T 和pGEM®-T Easy 載體連接時只能使用本系統(tǒng)提供的
Promega 公司T4 DNA連接酶。其他商用的T4 DNA連接酶可能具有外切
酶活力，造成載體的末端胸腺嘧啶被除去。
2． ２×快速連接緩沖液含有ATP，在溫度波動時ATP易降解?？蓪⒕彌_液分
裝成一次用量的小管，這樣可避免反復(fù)凍融及經(jīng)常的溫度變化。
3． 在每次使用２×快速連接緩沖液時一定要用混懸器混勻。
4． 長孵育時間可增加轉(zhuǎn)化子的數(shù)目，一般4℃孵育通宵可得到更多的轉(zhuǎn)化子。
5.一定要使用高效率的感受態(tài)細(xì)胞（1×108cfu/µg DNA）進(jìn)行轉(zhuǎn)化，建議使用JM109 高效感受態(tài)細(xì)胞（產(chǎn)品目錄號：L2001）
