說明 |
| 天然的和重組的RNasin® 核酸酶抑制劑具有廣譜的抑制RNase 活性的性質(zhì),包括抑制中性型(neutral type) 真核RNase。該抑制劑的分子量為50kDa,按1:1 的比例與RNase 非共價(jià)結(jié)合發(fā)揮抑制作用。RNasin® Ribonuclease Inhibitor 與RNase( 如RNase A) 結(jié)合的Ki 值大約是10-14M。Promega 提供兩種不同的抑制劑:天然的RNasin® Ribonuclease Inhibitor 和重組的RNasin® Ribonuclease Inhibitor。兩種產(chǎn)品均通過離子交換和親和層析相結(jié)合的純化方法純化得到。

RNases are ubiquitous, cause RNA degradation and can compromise RNA integrity. Recombinant RNasin® Inhibitor is a 50kDa protein that inhibits RNase A family and human placental RNases by noncovalently binding to RNases in a 1:1 ratio. Recombinant RNasin® Inhibitor does not inhibit RNase T1, S1 nuclease, RNase from Aspergillus, RNase H, RNase ONE™ Ribonuclease and enzymes for downstream applications such as GoScript™ Reverse Transcriptase, AMV/M-MLV reverse transcriptases, SP6, T7/T3 RNA polymerase, and Taq DNA polymerases. Broad-Spectrum RNase Inhibition: Inhibits common eukaryotic RNases.
Compatible: Does not inhibit SP6, T7 or T3 RNA Polymerase; GoScript™ Reverse Transcriptase, AMV or M-MLV Reverse Transcriptase; or Taq DNA polymerase.
Flexible: Functional across wide pH range (pH 5–8) enabling use in many downstream assays.
Recombinant: Made in bacteria, minimizing the chances of human nucleic acid contamination.

One unit is defined as the amount of RNasin® Ribonuclease Inhibitor required to inhibit the activity of 5ng of ribonuclease A by 50%. Activity is measured by the inhibition of hydrolysis of cytidine 2,3´-cyclic monophosphate by ribonuclease A.
