固定/ 破膜劑

（一）應(yīng)用于流式細(xì)胞儀檢測臨床全血標(biāo)本細(xì)胞內(nèi)多項(xiàng)免疫學(xué)指標(biāo)
肝素鈉抗凝的全血樣本，室溫存放，在24 小時(shí)內(nèi)使用。
1. 取抗凝血50ul 放入流式管中。
2. 加入檢測細(xì)胞表面分子用的熒光素標(biāo)記抗體，混和均勻，室溫避光孵育20 分鐘 。
3. 加入1ml 溶血素，振蕩均勻，室溫放置20 分鐘，離心（300g 5 分鐘）去除上清液。
4. 用1ml PBS 重懸洗滌細(xì)胞，離心（300g 5 分鐘）去除上清液。
5. 加入300ul 固定/ 破膜劑，振蕩均勻，室溫放置20 分鐘。離心（300g 5 分鐘）去除上清液。
6. 加入胞內(nèi)細(xì)胞因子檢測用熒光素標(biāo)記抗體，混和均勻，室溫避光孵育20 分鐘 。
7. 用1ml PBS 重懸洗滌細(xì)胞，離心（300g 5 分鐘）去除上清液。
8. 上機(jī)分析。

（二）應(yīng)用于流式細(xì)胞儀檢測外周血PBMC 的細(xì)胞內(nèi)多項(xiàng)免疫學(xué)指標(biāo)
1. 使用淋巴細(xì)胞分離液分離外周血樣本，獲得新鮮PBMC（外周血單個核細(xì)胞）后，取1X106的細(xì)胞放入流式管中。
2. 加入檢測細(xì)胞表面分子用的熒光素標(biāo)記抗體，混和均勻，室溫避光孵育20 分鐘 。
3. 加入300ul 固定/ 破膜劑，振蕩均勻，室溫放置20 分鐘。離心（300g 5 分鐘）去除上清液。
4. 加入檢測細(xì)胞內(nèi)細(xì)胞因子用的熒光素標(biāo)記抗體，混和均勻，室溫避光孵育20 分鐘 。
5. 用1ml PBS 重懸洗滌細(xì)胞，離心（300g 5 分鐘）去除上清液。
6. 上機(jī)分析。
