1.通常細胞增殖實驗每孔加入100微升2000個細胞，細胞毒性實驗每孔加入100微升5000個細胞(具體每孔所用的細胞的數(shù)目，需根據(jù)細胞的大小、細胞增殖速度的快慢等因素決定)。按照實驗需要，進行培養(yǎng)并給予藥物刺激。

推薦操作：通常建議培養(yǎng)板邊孔不用于實驗，僅加入100μL的培養(yǎng)基以減少培養(yǎng)過程中蒸發(fā)的影響

2.每孔加入10微升增強型CCK-8溶液。如果起始的培養(yǎng)體積為200微升，則需加入20微升增強型CCK-8溶液，其它情況以此類推?？梢杂眉恿讼鄳?yīng)量細胞培養(yǎng)液和增強型CCK-8溶液但沒有加入細胞的孔作為空白對照。如果擔(dān)心所使用的藥物會干擾檢測，需設(shè)置加了相應(yīng)量細胞培養(yǎng)液、藥物和增強型CCK-8溶液但沒有加入細胞的孔作為空白對照。

推薦操作：檢測前，用新鮮的培養(yǎng)基或不含血清的培養(yǎng)將CCK-8按1:10（CCK-8試劑：培養(yǎng)基）稀釋，混勻配成CCK-8工作液。然后吸去培養(yǎng)孔中的培養(yǎng)上清，加入稀釋好的CCK-8工作液，100μL/孔。此操作可減少重復(fù)樣本間的誤差。

3.在細胞培養(yǎng)箱內(nèi)繼續(xù)孵育1-4小時，對于大多數(shù)情況孵育1小時就可以了。時間的長短根據(jù)細胞的類型和細胞的密度等實驗情況而定，初次實驗時可以在0.5、1、2和4小時后分別用酶標(biāo)儀檢測，然后選取吸光度范圍比較適宜的一個時間點用于后續(xù)實驗。

4.在450nm測定吸光度?？梢允褂么笥?00nm的波長，例如650nm，作為參考波長進行雙波長測定。
