elisa技術(shù)流程
ELISA的基礎(chǔ)是抗原或抗體的固相化及抗原或抗體的酶標記。結(jié)合在固相載體表面的抗原或抗體仍保持其免疫學活性，酶標記的抗原或抗體既保留其免疫學活性，又保留酶的活性。在測定時，受檢標本（測定其中的抗體或抗原）與固相載體表面的抗原或抗體起反應(yīng)。用洗滌的方法使固相載體上形成的抗原抗體復(fù)合物與液體中的其他物質(zhì)分開。再加入酶標記的抗原或抗體，也通過反應(yīng)而結(jié)合在固相載體上。此時固相上的酶量與標本中受檢物質(zhì)的量呈一定的比例。加入酶反應(yīng)的底物后，底物被酶催化成為有色產(chǎn)物，產(chǎn)物的量與標本中受檢物質(zhì)的量直接相關(guān)，故可根據(jù)呈色的深淺進行定性或定量分析。由于酶的催化效率很高，間接地放大了免疫反應(yīng)的結(jié)果，使測定方法達到很高的敏感度。 ELISA可用于測定抗原，也可用于測定抗體。
然而，影響Elisa試驗結(jié)果的因素很多，故加強各個環(huán)節(jié)的質(zhì)量保證才能充分發(fā)揮其方法學的優(yōu)點。
試驗原理
試劑盒是固相夾心法酶聯(lián)免疫吸附實驗（ELISA）.已知濃度的標準品、未知濃度的樣品加入微孔酶標板內(nèi)進行檢測。先將生物素標記的抗體同時溫育。洗滌后，加入親和素標記過的HRP。再經(jīng)過溫育和洗滌，去除未結(jié)合的酶結(jié)合物，然后加入底物A、B，和酶結(jié)合物同時作用。產(chǎn)生顏色。顏色的深淺和樣品中的濃度呈比例關(guān)系。
原理
免疫測定技術(shù)的基礎(chǔ)在于抗原抗體之間的特異結(jié)合反應(yīng)，所以任何優(yōu)質(zhì)的診斷試劑離不開優(yōu)質(zhì)的原料，如抗原抗體，酶等。以前用于免疫測定的抗原通常為各種純化抗原，而抗體則為純化抗原免疫動物后獲得的多克隆抗體和使用雜交瘤技術(shù)得到的單克隆抗體，而抗原或抗體的標記物如酶標結(jié)合物則通過各種化學合成方法制備。近年來，隨著分子生物學的發(fā)展，使用基因工程方法制備各種特殊的抗原或抗體及其酶結(jié)合物等免疫測定試劑幾成為現(xiàn)實的新一代試劑，各種新型使用的測定方法也不斷出現(xiàn)。
HBsAg試劑特點（檢測模式：臨床抗體夾心法）：包被抗體為山羊多抗。多抗具有高親和性和對抗原各種表位的反應(yīng)性。酶表抗體為與HBsAg有不同結(jié)合位點的鼠復(fù)合單位，可測得HBsAg變異樣品，提高檢測的特異性（99.98%）提高檢測的靈敏度，應(yīng)用Parl Ehrich 學說（PEI）HBsAg標準品，對ad標準品的靈敏度為0.05ng/ml對ay標準品靈敏度為0.025ng/ml
ELISA檢測試劑盒應(yīng)用定性夾心免疫檢測技術(shù)，用合成的HEV多肽抗原包被微孔板板條，這些多肽是中國型HEV毒株核心氨基酸序列中抗原性很強的肽段，分別來自于該毒株的開放閱讀框2和開放閱讀框3。將樣品或標準品加入孔中并孵育，如果其中存在HEV IgM抗體，這些抗體就會與HEV 的多肽抗原結(jié)合，并固定在上面，洗板除去其它非特異性抗體和樣品中的其它成份。然后加入羊抗人IgM-HRP（辣根過氧化物酶）酶結(jié)合物，經(jīng)第二次孵育后，酶結(jié)合物就會與*次孵育結(jié)合上的HEV IgM抗體相結(jié)合，洗板除去未結(jié)合的酶結(jié)合物，加入TMB底物溶液，在第三次孵育時會發(fā)生酶-底物反應(yīng)，只有那些含有HEV IgM抗體和酶結(jié)合物所形成的復(fù)合物的孔才會發(fā)生顏色變化，加入硫酸溶液終止酶和底物間的反應(yīng)，并在波長: 450nm處測量O.D.值,按照本HEV IgM抗體elisa試劑盒的測試標準, O.D.值大于或等于Cut-Off值的樣品被認為是初試陽性。
